HCS
Toxic, Category 3 Carcinogen

FIRST-AID MEASURES EYES: Flush with water for 15 minutes. Seek medical advice if irritation persists.
SKIN: Flush with water, then wash thoroughly with soap and water. Remove contaminated clothing and wash
before reuse. Seek medical attention if irritation persists.
INHALATION: Remove the victim from exposure and move to fresh air. If breathing is difficult, give oxygen. If
not breathing, give artificial respiration. Keep victim quiet and warm. Seek immediate medical attention.
INGESTION: Drink water and seek immediate medical attention. Avoid alcoholic beverages. Never give
anything by mouth to an unconscious person. DO NOT induce vomiting.

FIRE-FIGHTING INFORMATION Use media suitable to extinguish the supporting or surrounding fire. Wear NIOSH (or equivalent) approved self
contained breathing apparatus. For small fires only: use carbon dioxide, dry powder or foam. This material
decomposes at temperatures above 185掳C emitting toxic fumes of carbon and nitrogen oxides.
Flash Point = No data available.

ACCIDENTAL RELEASE MEASURES Wear appropriate personal protective equipment and clothing including lab coat, safety goggles, gloves and NIOSH-
approved respirator. Collect in a manner that does not create dust and place in a suitable waste container. Avoid
contact of material with skin or eyes. Use adequate ventilation.

HANDLING AND STORAGE Wear appropriate personal protective equipment and clothing including lab coat, safety goggles, gloves and NIOSH-
approved respirator. Avoid contact of material with skin or eyes. Use adequate ventilation. Light sensitive.
Hygroscopic. Store at +4掳C away from incompatible materials.

PERSONAL PROTECTION Wear appropriate personal protective equipment and clothing including lab coat, safety goggles, gloves and NIOSH-
approved respirator. A qualified industrial hygienist should evaluate the need for respiratory protection. Use
respiratory protection approved by NIOSH (or equivalent) and appropriate to the hazard. Avoid contact of material
with skin or eyes. Mechanical ventilation or local exhaust as needed to control exposure to dust, vapors or mists.
Access to a safety shower and eye-wash. Wash thoroughly after handling.

PHYSICAL AND CHEMICAL Appearance: Bright yellow, orange crystalline powder Boiling Point: No data available
PROPERTIES Vapor Pressure: No data available Vapor Density: No data available

MSDS information may be continued on back of page.
STABILITY AND REACTIVITY Product is stable. Hygroscopic. May decompose on exposure to light. Protect from light and moisture. Hazardous
decomposition products include carbon and nitrogen oxides. Incompatible with strong oxidizing agents and strong
acids. Hazardous polymerization will not occur.

TOXICOLOGICAL INFORMATION EFFECTS OF OVEREXPOSURE:
EYES: Contact may cause irritation.
SKIN: May be toxic if absorbed through the skin and possibly lead to symptoms described under ingestion.
Contact may cause irritation and dermatitis.
INHALATION: May be toxic by inhalation and possibly lead to symptoms described under ingestion. May
cause irritation to mucous membranes and upper respiratory tract.
INGESTION: Toxic if swallowed. Chronic ingestion or excessive dosage may cause leukopenia, anemia,
thrombocytopenia, nausea, vomiting, diarrhea, dizziness, headache, fever, ulcerative stomatitis, necrosis of
the interdental papillae, hepatitis, pneumonitis, mucositis, renal failure, dark blood stool, blurred vision,
decreased resistance to infection and hair loss. Methotrexate is both teratogenic and a Category 2
mutagen causing DNA damage. Chronic exposure may cause mylelosuppresion and liver and kidney
damage. Pregnant women should avoid contact with Methotrexate due to the possibility of fetal
deformation or fetal death. May cause harm to the unborn child. May cause heritable genetic damage.
NOTE TO PHYSICIAN: Calcium leucovorin is a potent agent for neutralizing the immediate effects of
methotrexate on the blood-forming system. In general, when overexposure is known or suspected,
calcium leucovorin should be administered as soon after exposure as possible. See the latest edition of the
Physicians Desk Reference for recommended doses and routes of administration. Methotrexate blood levels
should be monitored and the calcium leucovorin dose adjusted accordingly.
TARGET ORGAN(S): Blood, Bone Marrow, Liver, G.I. System, Kidneys, Immune System and Lungs.
ADDITIONAL INFORMATION:
Methotrexate is a cytotoxic agent specifically designed to kill cells by preventing division or inhibiting cell growth.
The cell damage from cytotoxic agents is accumulative with a build-up of effects on continuing exposure
(irrespective of whether the drug itself is accumulating in the body).
Reproductive effects, irritation, mutation, tumorigenic and toxicity data listed in RTECS under MA1225000.
Only select RTECS information is provided here. Please see actual RTECS entry for complete information.
TDLo Oral Human (man ) = 4256 ug/kg/2.7Y-I (1982). Oral Rat LD50 = 135 mg/kg (1982). Toxic effects may
included fibrosis, focal (pneumoconiosis), respiratory obstruction and aplastic anemia.

fetal death (1975).
Specific developmental abnormalities - musculoskeletal, craniofacial, central nervous
system, body wall, eye/ear and cytological (1968).

of implants) (1968) and abortion (1973).
Reproductive - Paternal effects - spermatogenesis (inc. genetic material, sperm morphology,
motility, and count) (1969). Maternal effects - ogenesis (1968).
Carcinogenicity data: NCI Carcinogenesis Studies (ipr): no evidence: mouse, rat.
